Also Anita,
I'm not sure what kind of exposure times you are seeing and where/when
in the stack you start seeing photobleaching but you can also look at a
more sensitive acquisition camera (assuming you are utilizing a CCD
camera) such as the new genre of intensified and/or back-illuminated
CCDs.
You've certainly received some excellent advice for which the research
is sure to keep you busy through the holidays.
Happy Holidays Everyone!

Dear Anita,
There may be a very simple solution to reduce the time without reducing
the z resolution. However I'm uncertain if it will meet all your
experimental aims. From your email it sounds like you are taking a full
z-series (ie. xyz data set). Would it suit your experimental aims to
collect just a vertical section (xz)? This would be much quicker to
collect and you would end up with just one image per sample, rather than
a z-series. Hence analysis would be a lot quicker.
