Yo estoy tratando de resolver un problema parecido y creo que una buena solución al tema es utilizar una fibra óptica para fotolisar tus compuestos.
Este esquema lo utiliza un amigo (Ariel Escobar) y fotolisa desde fuera del camino óptico con una fibra montada en un micromanipulador para aproximarla a la célula dentro del baño.
Es lo que yo voy a intentar.
El problema es que Ariel utiliza un laser UV (Escobar et al., Kinetic properties of DM-nitrophen and calcium indicators: rapid transient, response to flash photolysis Pflugers Arch. 1997 Sep;434(5):615-31). No sabemos como puede terminar siendo la eficiencia del flash con lámpara a través de la fibra óptica, pero vale la pena intentarlo. Hasta donde yo discutí con el habría que ser muy cuidadoso en la óptica a utilizar para introducir la luz de la lámpara en la fibra óptica.

Hi all,
I would like to collect opinions (specially from "hard-core" users) regarding
my problem. I use a BioRad 1024 with an inverted Nikon Eclipse. When I
insert the dicroic mirror of 400 nm (used to excite with UV from the
conventional Hg lamp) I still receive images when scanning with the 488
Argon laser. I want to adapt a UV lamp in the epifluorescent path to use
caged compounds while scanning fuo-4 loaded cells using that dicroic. My
question is, will these images be confocal? how will them be affected by
the presence of the dicroic?.
