Hi Doug,
Since a water pipe burst in the ceiling above the confocal in the Ophthalmology dept,, we are providing confocal service until their system is repaired.  I just spoke with someone imaging a whole mouse retina, he was using a 60X/1.35 oil immersion lens with a retina mounted in Vectashield,and  routinely images to ~150 um depth.  I can't say whether that is the optimal preparation, but its a leading lab in the field.   Vectashield is essentially glycerol, so that may be a good place to start.
