--What tissue? In neurons, ethidium bromide marks the cytoplasm well. In many
other cells, it's a nuclear stain. It can be excited well at 488 nm, 514 nm,
and 543 nm; 568 nm will also work if you want to avoid exciting fluorescein.
Its emission is orange/red and should be distinct from fluorescein, as long as
you monitor fluorescein emission at wavelengths shorter than about 550 nm.
Good Luck!
