Jessica,

I strongly urge you to follow the calibration route, using a stage
micrometer.  It will automatically take into account all the variations in
magnification for each objective as well as tube lengths and a variety of
camera factors (chip size, pixel size, etc.). You can use the stage
micrometer to calibrate any imaging software so that you can add an
appropriate micron bar to your images.  This approach is good science and
presentation methodology.
