We routinely use H&E stained sections for testing on our confocals.  They are flat,  fluorescent, and high contrast for transmission detectors.  (We never tested color imaging by serial transmission images with different lasers, but it should work, but why you'd want to do it this way?)

I had some fun a few years ago with various cross subtractions of H&E taken with different spectral ranges with widefield fluorescence, but whereas the end products looked cool, I don’t think they were any better for identifying the tumors in the tissue from the normal tissue than other methods.

Am presently designing a research that will require the use of microscope. But am not sure if I can visualise a paraffin embedded H & E stained tissues with confocal microscope.
