Has there been any validation experiment done to determine if there is
compression of thick tissue specimens (100-200 microns) during confocal
imaging with oil-immersion lenses (40/60/100X)?  I am imaging spinal cord
sections of 150 microns, and the neurons in them are distributed all across
the thickness of a section.  I am interested in determining if the estimates
of thickness that I get for neurons on one side of the section are affected
by any compression of the tissue by the advancing objective during data
collection.
 
Any input is greatly appreciated.
