I can't comment on the dye you're proposing, but there is a published
method using 488nm excitation of SNARF read at its isosbestic point of
around 620nm (this dye is in itself a pH-dependent ratio emitter, but the
isosbestic point doesn't change in intensity), the reference is
"Improved method for Measuring Intracellular Ca++ with Fluo-3"G.T.
Rijkers,et al. Cytometry 11, 923 (1990).
The readout was flow cytometric, but should adapt to your setup.
