Hello everyone,

I purchased the DiO cell labeling kit from Molecular Probes to label the

membranes of fixed Caco-2 cells. I followed the manual for the staining
process and used different incubation times. Unfortunately I couldn't
get a uniform staining of all cells and the specimen were bleached with
a regular epifluorescence microscope within one minute. Did anyone else
have similar problems with Caco-2 cells or other cell lines? Since I
want to investigate these specimen with our confocal microscope I can't
afford any bleaching in that amount. Of course I could use another probe

(any suggestions?), but it is really annoying to buy a cell labeling kit

that doesn't even deliver satisfactory results. So please let me know if

you have any similar experiences concerning this kit.
