I assume you want to label cell nuclei and not DNA in vitro. For both purposes Molecular Probes has a series of fluorophores that bind to nucleic acid. Some are intended to be membrane permeant and are useful for live cells, others are meant to be used on fixed cells. I have used one of the "membrane impermeant" labels (YO-PRO-1, using 488 laser excitation and FITC filter set) in live tissue and had very good label. There are many others to choose from.

Subject: DNA staining

Hi everyone,
I am looking for staining the DNA of a mouse ovocyte. It is for dual
labeling and my 1st fluorochrome is Cy3. Propiduim iodine and Ethiduim
bromide are in the range of Cy3 so I can't use them. I cannot use UV's
suitable fluorochrome because I don't have the laser for it. So I am
looking for DNA markers emitting in the range of FiTC or Cy5 for dual
labeling.
Can't you help me...
Thanks
Louis Villeneuve
