If you want to calibrate your system, there is an article on calibration
of confocal systems by Lerner et al.:

Hi all,

I have spent a couple hours "google"ing and reviewing manuals/pdfs provided
by Zeiss without success to locate the following:

What are the units for the emission intensity graphs (and tables) one can
create when selecting an ROI within a  lambda stack?  When the graph data
are not normalized, the Y axis for intensity appears to read from 0-250
(with an 8 bit image - I've not taken any 12 bit yet), yet no unit is given.
Does this mean that it is reading brightness on a scale of
0-256 like an 8 bit digital cameras would and if so, what is the proper
technical label for this attribute?
