Hi all!

A student here is working on the 3D structure of paper of different 
kinds. The main purpose is to get images of the intact fibre structure 
to enable thresholding and subsequent measurement of length and 
directions of the fibres. Fine structure of the fibres is not critical 
but it is very important to avoid treatments that would cause swelling 
(e.g. water soluble stains).

I would be very glad for any suggestions on suitable stains, 
embeddings, or other treatments. It is necessary capture a large volume 
so something that will reduce opacity is a must.
We primarily intend to use the confocal where we have 457, 476, 488, 
514, 568, and 633 nm laser lines, but UV excitation on a wide field 
microscope with subsequent deconvolution is also a possibility.

Serial section is of course possible, but there is quite a large number 
of samples so it would not be practical to do it that way.
