Many users here have mentioned that the Confocal Image collected with
the 405 does not represent the image (conventional - ie Eyeball) viewed
using the standard UV filter set.
I do not have experience with this system doing (un)caging type
experiments, BUT the Olympus folks have been using the 405 for such
applications for a couple years now on the FV1000.

It works great. Very stable emission (intensity) from what I've
observed on our AOBS SP2.
The 405nm isn't a true UV but works very well for DAPI and other normal
UV excited dyes. It seems to have less of a bleaching affect on other
fluorochromes than the 364 on our Microcosm powered LSM410.
I would recommend considering changing the conventional fluorescent
filters from the standard DAPI set to one that would match the 405
laser, for example Chroma set 11005v2

Hi Everyone,
We are purchasing (unless someone talks me out of it really quick) a 405
upgrade to our Leica SP2 AOBS system primarily for imaging DAPI and
Hoechst. I would like to hear your comments on two things. Any
positive or negative aspects you have encountered with these lasers and
what additional applications (uncaging etc)for which you have
successfully used this laser.
