One of our clients wants to look at the internal structure of bone biopses
from deer rib and pelvis. The biopsy will be 3mm diameter and 3mm long and
then cut down the middle longitudinally. The idea will be to focus past
the damaged cut surface to look at the structure of the trabeculae, measure
thickness and estimate relative volumes.
Is there a non-specific fluoro stain for bone? Can it be injected
in vivo for uptake (presumably this would have to work on a localised basis
for deer)? Would the 1.5mm thick sections have to be further sectioned?
What would be the best sample preparation?
Tetracycline will be injected in vivo to indicate new bone
formation. What are the excitation/emission spectra (presumably 568nm
would excite)? Would it be possible to double label with the non-specific
label -and what about the crossover?
Any help would be greatly appreciated, particularly as bone is a new tissue
for us.
