Dear confocalists,
 
Does anyone know of difficulties in loading intact intestinal crypts
(mouse) with fluo-3 or fura-red (both AM)? In my crypt preparations
(isolated using an EDTA protocol) I don't get fluo-3 signals, and
fura-red signals only in cells located at the base of the crypt. I
have tried different concentrations, loading times and temperatures,
all with no result. Moreover, loading cells with SNARF-AM results in
a bright and evenly distributed fluoresence in all cells of the
crypts.
 
Any help is appreciated!
