Re> Andrew Vaillants question concerning CY5 imaging in Leica CLSM: Andrew, there are two options for triple labelled specimens in the Leica CLSM: 1. You get a triple dichroic 488/568/647 as main beam splitter and a longpass filter 665 as CY5 barrier filter. This allows for sequential and simultaneous imaging and certainly is the most flexible choice. The drawback is a sligthly reduced efficieny for green and red emission [not deep red CY5 emission] due to the necessity to have barrier filters of appropriate bandwidths to get no reflections of the higher lines into the image. 2. For pure sequential imaging, we can provide a 660nm dichroic with a longpass 665 nm for the CY 5 detection. Both filter combinations are standard Leica parts.
