I'm quite aware of the fact that the sensitivity of the eye dcereases repidly at longer wavelength. Those weak signals of Phalloidin 633 we measured either by using the PMT-s of our Zeiss 510 LSM or by using a cooled CCD camera (we tried a Kappa, and a Till Imago) (in this latter cases of course we used epi-fluorscence illumination either with the help of a laser or a HBO lamp or a Till monochromator) and in all cases the signal stayed weak compared to the shorter wavelength. Of course it is possible that there is a camera which we haven't tried and with which one could see those signals.... However so far we had never problems with visualizing Cy5 signals.
