Subject: Optimum well medium volume for imaging in 96 wells plate

Hi all,
 
We would like to get best  confocal images (on an Olympus FV300 confocal) of dim signal live cells cultured in a 96 well plate (glass bottom and dark edges).  Does some of you have found what is the best volume of medium to put per well in order to get a wide area of good focus for a couple of hours (2 to 3 hours)?  Any hints would be greatly appreciated.  Thanks a lot!
