Rolland,
if your microscope is of the fibre-coupled type and if , if you measure
over prolonged time(hours), the signal actually fluctuates (oscillates)
rather than steadily goes up, you are dealing with a phenomenon that has
to do with polarization effects on the dichroics/AOTF.
In that case, check the emaillist of about one month ago where we
discussed this in extenso. No cure yet.
Otherwise, pray, or refocus your research to running old fasioned gels
again.
