I am experiencing problems with GFP and LacZ detection. After transplantation in cardiac tissue, GFP positive cells do not appear under the fluourescent filter while the cells before transplantation were very bright under the same filter. With LacZ, the histochemical staining is not working for the same. The LacZ labelled cells after even 1 week of transplantation are not appearing in the tissue. I am using the standard protocols for fixation and staining and also tried different methods but so far did not get positive results. Any suggestions in this regard is highly appreciated
