We have used PI frequently for delineating dead cells in confocal
studies of living tissue (cardiac myocytes) by including PI (about 5
micromol.) in the superfusion buffer. Detect in red channel (exc 568,
em at 640). Remember that in living cells, PI does not cross the
plasma membrane unless the membrane is damaged - I have no
experience with fixed tissue.
 
PI binds to DNA, so the nucleus is almost uniformly stained in
interphase. I don't know if others are looking at nuclear cycles with PI
- might be interesting to hear about this.
