IMHO the only way to discover which software you need for your use is to
try it, talk to the sales people, look at the manual, etc. In other
words, each one of us has to do our own "dirty work" to find the
application specific to our own needs. We have four packages on our campus
and they all are owned by different groups with their own applications,
and each group likes what they have.

It IS clear from the comments & warnings given that there are differences in expense, ease of use & learning curve. But in discussing the differences in ACTUAL CAPABILITIES of the programs, what exactly are the differences, how do they show up, under which types of use do they become apparent? Specifics, please!

To make up for this it would be helpful if you could provide supporting details for your own observations, to let the audience keep your comments in perspective. For example, one would EXPECT an image analysis program to do (2D) thresholding and morphometry. Is UI different in these functions than other programs ? (perhaps specifically in comparison to the competitors in the roughly similar price range)? HOW? What extra capabilities in Imaris make it worth the extra cost? What functions should we (ignorant newcomers looking to buy our first or second program) be aware of that the lower price programs LACK but that we may find essential for a particular use?
