For imaginal disks I used 0.1 mg/ml in PBS for ~ 90 min at 37 degree (after 5
min in 1%Triton in PBS). If you test a new tissue, I would try e.g. 30 min, 60
min, 90 min and 120 min and just check the PI-stained result under the
microscope. Hope this helps
