Hi Steve,

Calcofluor is used to stain cellulosic cell walls. It is best used with fresh
sections cut either by hand or by cryotome, although it may also be used
effectively with dewaxed sections cut from wax-embedded blocks. Calcofluor
White M2R (American Cyanamid, Sigma) is used at concentrations of 0.1 mg/ml
(aqueous) for 30 sec to 5 min. After water wash, sections are mounted in water
or aqueous mountant and viewed with UV excitation. The pale blue signal is
generally very strong and anti-fade reagents may not be required.

Are there any recommendations out there for a fluorochrome suitable for
cellulose fibers in plant cell walls, alive and/or fixed?.
