Dear Dr. Taylor,
I can recommend you the dye congo red which can be excited with the 488 or
514 nm laser lines. The dichroic properties of the dye can be used to
visualize the mean cellulose microfibril orientation in plant cell walls
as discussed in
Verbelen and Stickens (1995) In vivo determination of fibril orientation
in plant cell walls with polarization CLSM. J Microsc. 177, 1-6
Verbelen and Kerstens (2000) Polarization confocal microscopy and congo
red fluorescence: a simple and rapid method to determine the mean
