Dear Henna,

We do a lot of multipoint imaging with oil objectives on a number of
systems: ZeissLSM510, Leica SP2, DeltaVision (and also on a Zeiss based
wide-filed system. We have absolutely no problems to image 10-20 (or
even more locations) as long the sample is not larger then a ~ 1/2
object slide. We have the cells in LabTek or Ibidi chambers (or in a
home made holder with a 24mm cover slip as bottom). We have basically no
problems with the oil (or with temperature fluctuations). I think the
"secret" is, that (like it was suggested many time already on this list)
we have large box-type incubator enclosing the X-Y stage and a large
part of the microscope. We use different type of "boxes" (Solent, Life
Imaging Services or EMBL) - they all work fine. Another "trick" is to
check whether your cell-chamber is coupled to your stage in a
mechanically stable manner.

Cheers Gabor
