Fixation with pFA is not very reliable for fixing soluble cytoplasmic
proteins (a major proportion of which can still be washed out after
fixation) and probably dextrans also. Adding a detergent will probably
make it even worse. Adding a crosslinking fixative, such as
glutaraldehyde or EGS should make the fixation much more reliable. And,
as Don has already pointed out, make sure that you use a FIXABLE dextran.
