Dear List,

I have obtained pRSET-B mcherry plasmid from another lab. With help
of biologist friend, I am trying to sub-clone mCherry into another
plasmid (pCDNA3) such that we can express fusion of our protein and
mCherry. How do people find mCherry fusions work
properly?Protein-mCherry or mCerry-protein and does it necessarily
need linker?

Also, I don't understand most of the terms in plasmid map (as of now I
have taken only one course in molecular biology). Could anyone please
help me expand them?

pUC - is it a promoter? If yes, can it be used to express mcherry
protein in colon cancer cells as a control for the fusion?

This is the link to the plasmid map:

Thanks very much,
regards,
Shalin
