Perhaps you could apply CyGEL to this issue?
This is a thermo-reversible hydrogel which transits to a gel from sol when
warmed to 22 deg C and above.
This will immobilise (with infinite reversibility) particles, organisms or
cells.
As a hydrogel, it provides the aqueous interface for the TIRF measurement
you need and has excellent optical properties - clear and with a
refractive index of 1.36, close to that of water.  It has no significant
autofluorescence.
