We have recently tried using some of the xylene-based mountants.
Cytoseal (VWR 97-8 catalog, pp. 852 and 1097) worked quite well in our
fibroblast samples. The media does come in two, viscosities: low for small
stuff (i.e., fibroblasts) and high for bulky cells (i.e., oocytes). The
probes do not seem to be affected by the brief alcohols and xylene
dehydration steps. This also eliminates using nail polish to seal the
coverslips.
