Udo
I just carried out an experiment comparing images with a x63 and
x100 objective on bacteria stained with nile blue and I have to admit
that images were not significantly different. Using the zoom with a
x63 lens does not result in a more "pixelated" image for the reasons
already described. Perhaps I have a natural suspicion about using a
zoom. I do, however, use the x100 quite a lot for optical microscopy
and would still recommend it for that purpose.
Thanks to you guys out there for correcting my rash statements -
Apologies, Udo, for causing any confusion. For confocal work only,
the x63 would be best, particularly since you will be looking at
microbial populations rather than individual cells.
