O.k., is there a formula for determining the depth of an optical sectioning depth?
I assume it will vary with the NA of the lens (and diameter of the pinhole).
Specifically I am working with a Nikon PCM2000 (Single Pinhole) on a 160mm Nikon
Optiphot2, with a 20um and a 50um pinhole. Direct measurment via fluorescent beads is
going to run into airy disk problems, and determination from physically sectioned
(microtomy sections) is limited by the 'accuracy' of the microtome, as well as the
number of various sections required.
