Hi,

We use 1 x PBS, 1% Triton X-100, 1% BSA as a buffer for all steps after 4%
Paraformaldehyde fixation in whole Drosophila brains. This works fine with
anti-betagalactocidase but might not be suitable for other antibodies.

I've also used 0.1% Tween 20 some time ago and it appeared to work fine.

As for using alcholos/xylene, its fine so long as there are no tissues
connected that have pigments in them (we have problems with fluorescent
pigments in the Drosophila retina).
