Dear Listmembers,
I would agree with Holly's suggestions. On a dichroic based SP2 you
will have to be a bit further away from the laser lines. I find on my old
SP1 (dichroic based) I need the polarizer to remove the reflection at the
center of the image.
If you are using an AOBS system, you can get quite close. This will depend
on the wavelength, so at 488 nm you can be about 2 nm away, while at 633 you
will need to be a bit further.
In either case make sure your laser power (AOTF) is not too high, otherwise
you will certainly get some interference patterns.
Cheers,
Owen
